The argument that automatic processes are responsible for affective/evaluative priming effects has been primarily based on studies that have manipulated the stimulus onset asynchrony (SOA; i.e., the interval between the onset of the prime and the onset of the target). Moreover, these SOA studies provide an insight in the time course of the activation processes underlying automatic affect/attitude activation. Based on a fine-grained manipulation of the SOA employing either the evaluative decision task (Experiment 1) and the pronunciation task (Experiment 2) (...) we concluded that affective priming, and hence automatic affect activation, is based on fast-acting automatic processes. The results of Experiment 3 provide a valid explanation for an apparent discrepancy between the results of Experiments 1 and 2 and previous findings. Finally, the results of Experiment 3 support the prediction of Jarvis and Petty (1996) that affective priming effects should be stronger for participants who are more chronically engaged in conscious evaluations. Understanding the functional mechanisms underlying genetic signals associated with complex traits and common diseases, such as cancer, diabetes and Alzheimer's disease, is a formidable challenge. Many genetic signals discovered through genome‐wide association studies map to non‐protein coding sequences, where their molecular consequences are difficult to evaluate. This article summarizes concepts for the systematic interpretation of non‐coding genetic signals using genome annotation data sets in different cellular systems. We outline strategies for the global analysis of multiple association intervals and the in‐depth (...) molecular investigation of individual intervals. We highlight experimental techniques to validate candidate (potential causal) regulatory variants, with a focus on novel genome‐editing techniques including CRISPR/Cas9. These approaches are also applicable to low‐frequency and rare variants, which have become increasingly important in genomic studies of complex traits and diseases. There is a pressing need to translate genetic signals into biological mechanisms, leading to prognostic, diagnostic and therapeutic advances. (shrink)

Depression is vastly heterogeneous in its symptoms, neuroimaging data, and treatment responses. As such, describing how it develops at the network level has been notoriously difficult. In an attempt to overcome this issue, a theoretical “negative prediction mechanism” is proposed. Here, eight key brain regions are connected in a transient, state-dependent, core network of pathological communication that could facilitate the development of depressive cognition. In the context of predictive processing, it is suggested that this mechanism is activated as a response (...) to negative/adverse stimuli in the external and/or internal environment that exceed a vulnerable individual’s capacity for cognitive appraisal. Specifically, repeated activation across this network is proposed to update an individual’s brain so that it increasingly predicts and reinforces negative experiences over time—pushing an individual at-risk for or suffering from depression deeper into mental illness. Within this, the negative prediction mechanism is poised to explain various aspects of prognostic outcome, describing how depression might ebb and flow over multiple timescales in a dynamically changing, complex environment. The effect of specificity of autobiographical memory (AM) retrieval on the affective impact of an emotional event was examined. In Study 1 (N = 90) the impact of a negative and positive experience was compared between student participants who habitually retrieve autobiographical memories (AMs) in a specific way and participants who generally retrieve less specific memories. In Study 2 (N = 48) the effect of an experimentally induced (specific vs. overgeneral) retrieval style on the impact of a negative experience was (...) studied in student participants who habitually retrieve less specific memories. Study 1 replicated the finding of Raes, Hermans, de Decker, Eelen, & Williams (Citation2003) that a negative event leads to less subjective distress in low‐specific participants as compared with high‐specific participants. However, both groups did not differ in their affective reaction to a positive event. Important, reduced memory specificity was associated with “repressive coping”, providing further evidence for the idea that reduced memory specificity is used as an avoidant or repressive‐defensive mechanism to regulate negative affect (CitationWilliams, 1996). In Study 2, participants who were induced to retrieve memories in an overgeneral way experienced more distress following a negative event as compared with participants who were induced to retrieve memories in a specific way. Results are discussed in the context of recent findings concerning AM specificity and emotion regulation (CitationPhilippot, Schaefer, & Herbette, 2003). After the moral framework of sustainable development, the focus on climate change appears to take a lead in the practice and theory of environmental education. Inherent in this perspective is an apocalyptic message: if we do not rapidly change our use of energy resources, we will severely harm the life conditions of our children and grandchildren. In this article we argue that environmental educators should liberate us from this highly instrumental dictate by taking their cue from our daily care for (...) our environment as revealed in child?s play in nature. Inherent in the interaction of the Homo ludens with nature is a spiral time experience. This time condition elicits a particular responsibility: rather than calculating which means are necessary to prevent future disasters, we should respond with care to the concentric circles of an open and indefinite future. Therefore, environmental education should not aim to prevent future disasters, but foster anticipating care for nature. (shrink)

Begründung von Offenbarungserkenntnis, seine Korrelationsmethode, d.h. die Zuordnung von philosophischen Fragen und theologischen Antworten, sowie die Bedeutung von Tillichs Christologie - in der Spannung zwischen absoluten Geltungsansprüchen und deren Relativierung - für den interreligiösen Dialog heute. John Rawls wordt algemeen beschouwd als een van de meest invloedrijke filosofen van de voorbije eeuw. In zijn befaamde boek 'A Theory of Justice' uit 1971 staat het principe van de rechtvaardigheid centraal dat hij beschouwt als de belangrijkste maatschappelijke waarde in de samenleving. Mensen zijn fundamenteel gelijkwaardig. Daarom is het voor Rawls evident dat een ethisch systeem zich richt tot iedereen. Natuurlijk beseft hij dat mensen niet gelijk zijn of beter gezegd, niet over dezelfde begaafdheden beschikken noch over dezelfde (...) startposities. Waar het bij hem op aan komt is de onvrijwillige achterstanden, dus achterstanden die niet gebaseerd zijn op vrijwillige keuze, weg te werken. Epigenetic and transcriptional variability contribute to the vast diversity of cellular and organismal phenotypes and are key in human health and disease. In this review, we describe different types, sources, and determinants of epigenetic and transcriptional variability, enabling cells and organisms to adapt and evolve to a changing environment. We highlight the latest research and hypotheses on how chromatin structure and the epigenome influence gene expression variability. Further, we provide an overview of challenges in the analysis of biological variability. An (...) improved understanding of the molecular mechanisms underlying epigenetic and transcriptional variability, at both the intra- and inter-individual level, provides great opportunity for disease prevention, better therapeutic approaches, and personalized medicine. Epigenetic and transcriptional variability mediate phenotypic plasticity, enabling adaptation to changing environments. In this review, we describe the sources of inter- and intra-individual variability and discuss epigenetic regulators of gene expression variability, including DNA methylation and chromatin structure. Understanding these molecular mechanisms will improve therapeutic approaches and personalized medicine. (shrink)

Der Aufsatz stellt zunächst das konstruktive Abstraktionsverfahren von Paul Lorenzen vor, das eine Bezugnahme auf Mengen vermeidet und daher gestattet, den Terminus ‚Menge‘ selbst als einen Abstraktor einzuführen. Es werden dann weiter häufig übersehene Konsequenzen des Abstraktionsverfahrens für die Rede von abstrakten und konkreten Gegenständen aufgezeigt. Schließlich werden die Inkonsistenzbeweisversuche von Geo Siegwart dargestellt und kritisch diskutiert. Dabei wird gezeigt, dass diese darauf beruhen, dass die besagten Konsequenzen nicht berücksichtigt werden.

In the first part of this essay, some of the strategies atheists use to claim undeserved privileges for their position are analyzed. In particular, the connection between evidentialism and the idea that atheism is the natural starting position (“presumption of atheism”, Anthony Flew) is discussed. In addition, the extent to which the elimination of agnosticism as the third option between theism and atheism contributes to the discursive privilege of atheism is examined (with Paul Cliteur). In the second part, evidentialism (...) is reconstructed as a context-dependent theory of justification and the (A) theism problem is situated in the context of action instead of the context of knowledge. This avoids one-sided distribution of the burden of justification, especially the atheistic attempt to to burden the theist with the full burden of justification and at the same time to free the atheist from all burdens of justification. In this way, the discourse on the (a) theism problem can be reconstructed without assigning undeserved privileges. (shrink)

“Neo-inclusivism” is explained and assessed as an approach to the problem of the conflicting claims to truth of different religions, with reference to inter alia John B. Cobb (Jr.), Gavin D'Costa and Paul Ingram. For the neo-inclusivist the truth of a religious tradition depends on its inclusivistic capacity, i.e. its capacity to assimilate other traditions. For ex ample, by being enriched and transformed through “radical openness” to other traditions, while remaining “committed” to her own tradition – so the neo-inclusivist (...) claims – the theology of the Christian can become “truly global”. Neo-inclusivism seemingly transcends the absolutist habits of inclusivists, i.e. to assess the religious other in view of criteria (the inclusivist's own religious tradition) with which the other does not identify himself or herself. However, the neo-inclusivist plays down the possible limits of inter religious openness and is overly optimistic about the possibility of a “truly global” theology. Moreover, whether the neo-inclusivist's “committed openness” significantly differs from the approach of those who arbitrarily use the definitive norms of their own religious tradition to assess other traditions, is at least debatable. Whatever the case may be, insofar as tolerance cannot tolerate intolerance, the neo-inclusivist is necessarily intolerant, and, in this sense, “absolutist”, towards those who make their own beliefs normative for all and shun themselves from the assessments and new insights of others. Do adherents of different religious traditions communicate and, if so, how? What enables them to do so? What is interreligious “communication”? These issues are ad dressed with reference to Wilfred Cantwell Smith's hermeneutical rule, and to inter alia Paul Knitter, Martin Heidegger, Hans-Georg Gadamer, David Tracy, and John Dunne. Four responses to the question as to what permits interreligious communication are criticised. According to a fifth response, on which the author elaborates, interreligious communication is not – as the objectivist (...) claims – possible because members of different religious traditions speak the very same language, or – as the relativist claims – impossible because they speak radically different languages. Adherents of different religions communicate, if at all, by means of an analogical language. This language is the product of persistent efforts to understand the religion/faith of the other as he or she understands it. Paradoxically, the language that permits inter religious communication is thus the product of such communication. Without thereby reverting to objectivism or an essentialist view of religion, the author identifies five “less unstable” similarities between religions. These and other “less un stable” similarities might assist believers in their efforts to under stand the religious other. (shrink)

The Spirit (πνεῦμα) and peace (εἰρήνη) with God as opposed to the Flesh (σάρξ) and hostility (ἔχθρα) with God in Romans 8:6–8. A surprising number of exegetes do not address the question to what exactly εἰρήνη refers in Romans 8:6. The rest seem to be divided between interpreting it as an unspecified (eschatological) state of peace (šalôm), peace with one’s fellow humans, or peace with God. Based on the textual context, this article argues that the latter interpretative option is best. (...) In terms of metaphor, the relevant target domain here is that of the relationship with God, while the source domain reflected probably is that of diplomatic relations. In addition, peace (with God) here must be understood in relation to the rule of the Spirit, the decisive influence upon those who live κατὰ πνεῦμα (Rom 8:4–5). Antithetically, personified flesh rules over those who are ἐν σαρκί (Rom 8:8). This leads to a disposition of enmity toward God as well as the inability to submit to his law. For Paul being ruled by the Spirit, as a consequence of being reconciled with God through Christ (Rom 5:1, 10; 8:3–4), is a crucial aspect of being at peace with God. (shrink)

The rapid dynamics of COVID-19 calls for quick and effective tracking of virus transmission chains and early detection of outbreaks, especially in the “phase 2” of the pandemic, when lockdown and other restriction measures are progressively withdrawn, in order to avoid or minimize contagion resurgence. For this purpose, contact-tracing apps are being proposed for large scale adoption by many countries. A centralized approach, where data sensed by the app are all sent to a nation-wide server, raises concerns about citizens’ privacy (...) and needlessly strong digital surveillance, thus alerting us to the need to minimize personal data collection and avoiding location tracking. We advocate the conceptual advantage of a decentralized approach, where both contact and location data are collected exclusively in individual citizens’ “personal data stores”, to be shared separately and selectively, voluntarily, only when the citizen has tested positive for COVID-19, and with a privacy preserving level of granularity. This approach better protects the personal sphere of citizens and affords multiple benefits: it allows for detailed information gathering for infected people in a privacy-preserving fashion; and, in turn this enables both contact tracing, and, the early detection of outbreak hotspots on more finely-granulated geographic scale. The decentralized approach is also scalable to large populations, in that only the data of positive patients need be handled at a central level. Our recommendation is two-fold. First to extend existing decentralized architectures with a light touch, in order to manage the collection of location data locally on the device, and allow the user to share spatio-temporal aggregates—if and when they want and for specific aims—with health authorities, for instance. Second, we favour a longer-term pursuit of realizing a Personal Data Store vision, giving users the opportunity to contribute to collective good in the measure they want, enhancing self-awareness, and cultivating collective efforts for rebuilding society. The Monographs produced by the International Agency for Research on Cancer (IARC) apply rigorous procedures for the scientific review and evaluation of carcinogenic hazards by independent experts. The Preamble to the IARC Monographs, which outlines these procedures, was updated in 2019, following recommendations of a 2018 expert Advisory Group. This article presents the key features of the updated Preamble, a major milestone that will enable IARC to take advantage of recent scientific and procedural advances made during the 12 years since (...) the last Preamble amendments. The updated Preamble formalizes important developments already being pioneered in the Monographs Programme. These developments were taken forward in a clarified and strengthened process for identifying, reviewing, evaluating and integrating evidence to identify causes of human cancer. The advancements adopted include strengthening of systematic review methodologies; greater emphasis on mechanistic evidence, based on key characteristics of carcinogens; greater consideration of quality and informativeness in the critical evaluation of epidemiological studies, including their exposure assessment methods; improved harmonization of evaluation criteria for the different evidence streams; and a single-step process of integrating evidence on cancer in humans, cancer in experimental animals and mechanisms for reaching overall evaluations. In all, the updated Preamble underpins a stronger and more transparent method for the identification of carcinogenic hazards, the essential first step in cancer prevention. (shrink)

Activation of resting T lymphocytes through the T cell antigen receptor complex is initiated by critical phosphorylation and dephosphorylation events that regulate the function and interaction of a number of signaling molecules. Key elements in these reactions are members of the Src, Syk and Csk families of protein tyrosine kinases (PTKs) and the phosphotyrosine phosphatases (PTPases) that regulate and/or counteract them, such as CD45. The PTKs can autophosphorylate and phosphorylate each other at multiple sites and, as the result of these (...) interactions, they are induced to phosphorylate other cellular proteins. These phosphorylation events lead to modulation of enzymatic activities and/or serve as binding sites for other signaling molecules having phosphotyrosine‐binding Src homology 2 (SH2) domains. As a result, these proteins translocate to the receptor complexes and are juxtaposed to the kinases that phosphorylate them. Some of the SH2‐domain‐containing polypeptides lack enzymatic activities and, instead, serve as adapter molecules that couple the signal to downstream effectors, such as regulators of the Ras proteins, and further into serine/threonine‐specific protein kinase cascades. Through largely unknown steps these reactions lead to the transcription of previously silent genes, activation of lymphocyte effector functions, progression through the cell cycle and cell proliferation. The fifteen essays in this volume, rooted in the work of the Hellenistic Moral Philosophy and Early Christianity Section of the SBL, examine the works of Philodemus and how they illuminate the cultural context of early Christianity. Born in Gadara in Syria, Philodemus (ca. 110-40 BCE) was active in Italy as an Epicurean philosopher and poet. This volume comprises three parts; the first deals with Philodemus' works in their own terms, the second situates his thought within its larger Greco-Roman context, (...) and the third explores the implications of his work for understanding the earliest Christians, especially Paul. Die Kontroversen um Imre Lakatos' "Methodologie wissenschaftlicher Forschungsprogramme" haben zu der Frage geführt, ob auf Basis der von Lakatos entwickelten Standards für die Bewertung von konkurrierenden Theorien und Forschungsprogrammen auch heuristische Empfehlungen begründet werden können, die die von Wissenschaftlern auszuführenden Handlungen im Interesse der Maximierung von Erkenntnisfortschritt normieren. Eine solche Frage steht im Zusammenhang mit einem zentralen Anliegen der Popperschen Wissenschaftstheorie: "Wir wollen die Regeln, oder, wenn man will, die Normen aufstellen, nach denen sich der Forscher richtet, wenn er Wissenschaft (...) treibt, wie wir es uns denken". Ob die Methodologie wissenschaftlicher Forschungsprogramme, die Lakatos als Weiterentwicklung des kritisch-rationalistischen Falsifikationismus verstanden wissen will, diesen Ansprüchen genügt, wurde zuerst von Paul K. Feyerabend und Thomas S. Kuhn bezweifelt. Im folgenden werden zunächst deren Kritiken, die Antworten von Lakatos sowie Analysen von J. J. C. Smart, Wolfgang Stegmüller und Philip Quinn diskutiert. Anschließend wird ein neuer Versuch Alan Musgraves kritisch überprüft, unter Rückgriff auf die Bewertungskriterien der Methodologie wissenschaftlicher Forschungsprogramme heuristische Empfehlungen zu begründen. The present collection of essays, which originated as a colloquium at the Center for Hellenic Studies, starts, in the words of editor and organizer Dirk (...) Obbink, from the premise that "genre is a conceptual orienting device that suggests to the hearer the sort of receptorial conditions in which a fictive discourse might have been delivered." Obbink goes on to note that, as the discussion at the colloquium developed, "a more text-based conception of authorial performance" began to emerge (6). This evolution in turn parallels the movement in the volume from an initial discussion of archaic and classical Greek genres to those of Hellenistic poetry and its Roman imitators. One can only applaud this attempt to provide a more sophisticated framework for the reading and reception of ancient literature. As befits the stature of the sponsoring institution and of the many distinguished participants, all the contributions are of an unquestionable competence. In the end, however, it is unclear that we have achieved a more complex, refined, or problematized concept of genre than that with which we began. Indeed, with a few exceptions, most of the modern debates about the concept of genre are either ignored or cited in passing rather than directly engaged. Nonetheless, there are a number of interesting and important individual papers in this volume and only a few that disappoint.Among the most praiseworthy is Eric Csapo's "From Aristophanes to Menander? Genre Transformation in Greek Comedy." Csapo takes dead aim at the traditional narrative of the evolution from a politically engaged Old Comedy of ribald invective to an apolitical plot-centered New Comedy typified by Menander and his Roman epigones. Csapo's argument is that this narrative was manufactured, not through the accidents of preservation, but through a deliberate selection and canonization of texts by early Peripatetic critics and scholars. When we characterize Old Comedy as being typified by Aristophanes' carnivalesque assaults on the Athenian political establishment and then point to his plays as proof, we are not only engaging in circular reasoning, we are also treading a path that has been deliberately and tendentiously drawn for us. Thus, while the traditional narrative depicts a linear evolution from iambic to Old Comedy to New Comedy, which is portrayed as a movement from the personal and the topical to the universal and the apolitical, Aristotle himself, as Csapo notes (117), admits that Crates, Aristophanes' predecessor, abandoned the iambic and began to compose generalizing plots (Po. 1449b7-8). In short, plots of the kind deemed [End Page 119] typical of New Comedy are to be found at the very beginning of the comic form. By the same token, we have evidence of political comedy surviving well past Aristophanes' supposed invention of the Middle Comic style in the Plutus. Indeed, the last example of an Athenian comedy of political invective is Demetrius' Areopagite, dating from circa 294 B.C.E.Csapo's argument, of course, is not a simpleminded refusal to acknowledge change but rather an attempt to rethink the historical evolution of the comic genre outside the bounds of a narrow, teleological narrative. Genres are always complex negotiations between competing conceptions of the same form by different producers and consumers. At various times, for various intrinsic and extrinsic reasons, different versions of the same form will be ascendant or fading into eclipse. But it is never a question of an essence that either unfolds its fullness over time or declines from a single ideal instantiation.Less revolutionary but equally persuasive is Joseph W. Day's "Epigram and Reader: Generic Force as (Re-)Activation of Ritual." Day examines the relation between dedicatory epigrams on cult statues and their ritual contexts by arguing that the dedicatory genre is essentially performative rather than constative. Volume 1 of this biography of L. E. J. Brouwer was published in 1999.1 The volume under review here covers the period from the early nineteen twenties until Brouwer's death in 1966. It also includes a short epilogue that discusses the disposition of Brouwer's estate after his death, his influence on others, the paths of some of his students and colleagues, and other matters. Van Dalen notes in the Preface that in preparing this volume he consulted some historical studies that (...) appeared after the first volume was published. He also used new material from various archives. The biography contains interesting quotations from unpublished materials in the Brouwer Archive and from correspondence. The bibliographical references to Brouwer's publications, it should be noted, are somewhat different in this volume. This volume, like the first, contains some nice photographs and reproductions. I noted that there were many typographical errors in the earlier book but Volume 2 is relatively free of them.As is the case in Volume 1, the discussion of Brouwer's mathematical and philosophical work is woven into the narrative of Brouwer's life and times. The story in this volume starts with Brouwer's first contacts with Paul Alexandrov and Paul Urysohn in 1923. The interaction began when Urysohn announced that he had found a mistake in Brouwer's definition of dimension in Brouwer's 1913 paper on natural dimension. Was it just a slip of the pen, as Brouwer always maintained , or something more substantial? Urysohn and Alexandrov ultimately came to agree with Brouwer on the matter, and Urysohn was prepared to grant Brouwer priority for the definition of dimension. There were, however, ups and downs along the way. Karl Menger, through his own work in topology and dimension theory, soon got into the picture, and Brouwer and Menger were to …. (shrink)

An ongoing trend in technology policy has been to advocate participation. However, the author claims that lay citizens’ participation typically materializes in the form of a laboratory experiment at present. That is, lay participation as currently organized by professional participation experts under controlled conditions rarely is linked to public controversies, to the pursuit of political participation or to individual concerns. Derived from qualitative research on two citizen conferences, the author shows empirically that in practice, this laboratory participation leads to paradoxical (...) effects: successfully carrying out the experiment results in a systematic disappointment of the hope for gains in rationality typically attached to lay participation. Finally, the author relates this result to sociological debates about new modes of knowledge production. Under such a perspective, the author sees a paradoxical development: while society at large is becoming a laboratory in which knowledge is produced, participation practice is retreating from society into the lab.
